Quantitation of Human Cytomegalovirus(HCMV) by Enzyme Immunoassay

Abstract
The rapid quantitative method for the detection of HCMV is required to evaluate the clinical state for HCMV infections. We therefore attempted to develop enzyme-linked immunosorbent assay for the detection of HCMV in culture supernatant and urine
by
using monoclonal antibody(MCMVA 135) specific to HCMV glycoprotein B(gB). MCMVA 135 was purified by salting-out, and was attached to the plate. The culture supernatant of HCMV infected cells or urine from HCMV infected patient were applied as
antigens,
and then the peroxidase-conjugated MCMVA 135 was added. After the substrate was added the absorbance was measured at 405nm. The standard curve showed that HCMV in culture supernatant could be detected from 3¡¿10E3 PFU/ml to 5¡¿10E4 PFU/ml
linearly.
HCMV was not detected in all but one urine sample from eight healthy control persons whereas detected from at the concentration of 3¡¿10E3 PFU/ml to 5¡¿10E4 PFU/ml in all but one urine sample from eight HCMV-infected patients. These results
revealed
that the specificity and the sensitivity were 87%, respectively.
This detection method of HCMV antigen from the culture supernatant and urine would offer advantages in terms of quantitative sensitivity.
